- The polyhydroxyalkanoates (PHAs) are a group of polymers that accumulate in the bacterialcytoplasm forming inclusions called granules. These polymers, biosynthesized from removableresources, can be used as biodegradable plastics. Bacillus megaterium accumulates poly(3-hydroxybutyrate) PHB, a linear homopolymer of D(-)-3-hydroxybutyrate. The granule structureand the biochemical mechanisms involved in their synthesis are not completely understood. In thiswork a collection of B. megaterium mutants deficient in PHB synthesis was obtained. None ofthem showed PHA synthase activity. A system was developed for the study of heterologous genes,using these mutants and the Gram positive and negative bacterial shuttle plasmid pAT19, ascloning vector. In an attempt to understand the role of granule associated proteins and theirinteractions, this system was used to analyze if the 22 Mr protein of B. megaterium, that issupposed to perform the role of a structural protein, could be associated with PHA granulessynthesized by the Alcaligenes eutrophus synthase. The results showed an association to thegranules, whose synthesis was directed by an heterologous synthase.
