Evolución molecular de factores de transcripción del tipo zinc finger involucrados en el desarrollo de la arquitectura final de las gramíneas

RAMOSA1 (RA1) es uno de los genes seleccionados durante el proceso de domesticación del maíz y codifica para un factor de transcripción del tipo zinc finger. RAMOSA1 se caracteriza por tener en su secuencia un dominio de unión al ADN zinc finger del tipo C2H2 y dos motivos represores EAR. Ambos dominios represores son necesarios para la interacción de RA1 con el co-represor RAMOSA1 ENHANCER LOCUS2 (REL2). A través de la interacción RA1/REL2 se regula la expresión de genes específicos que controlan el destino y la identidad de los meristemas de rama durante el desarrollo de la inflorescencia de maíz. A pesar de la importancia de RA1 durante el desarrollo de la planta, se desconoce el origen, evolución y su función en otras especies. Por ello, el objetivo general de este trabajo fue estudiar RA1 desde una perspectiva evolutiva, identificar motivos conservados entre las proteínas y su correlación con la estructura secundaria y con los fenotipos generados en plantas de Arabidopsis que sobreexpresan RA1. Nuestros resultados indican que RA1 surgió a partir de dos duplicaciones consecutivas de SUPERMAN, dando lugar a tres linajes de secuencias de gramíneas: RA1-like A, RA1-like B y RA1. Por otro lado, analizamos los fenotipos observados en plantas de Arabidopsis que sobreexpresan RA1, SvRA1 y CeRA1 y se evaluó el potencial uso de la sobreexpresión de RA1 como herramienta biotecnológica. Como resultado, se observaron fenotipos de potencial valor innovador.
RAMOSA1 (RA1) is one of the genes selected during the domestication process of maize and encodes a zinc finger-type transcription factor. RA1 is characterized by a C2H2-type zinc finger DNA-binding domain and two EAR repressor motifs in its sequence. Both repressor motifs are required for the interaction between RA1 and the co-repressor RAMOSA1 ENHANCER LOCUS2 (REL2). Through the RA1/REL2 interaction, the expression of specific genes is regulated, controlling the fate and identity of branch meristems during maize inflorescence development. Despite the importance of RA1 in plant development, its origin, evolution, and function in other species remain unknown. Therefore, the main objective of this work was to study RA1 from an evolutionary perspective, identify conserved protein motifs, and analyze their correlation with secondary structure and the phenotypes observed in Arabidopsis plants overexpressing RA1. Our results indicate that RA1 originated from two consecutive duplications of SUPERMAN, giving rise to three lineages of grass sequences: RA1-like A, RA1-like B, and RA1. In addition, we analyzed the phenotypes observed in Arabidopsis plants overexpressing RA1, SvRA1, and CeRA1, and evaluated the potential use of RA1 overexpression as a biotechnological tool. As a result, phenotypes with potential innovative value were observed.
