El objetivo de esta tesis fue caracterizar a la población de linfocitos Tγδ luego de la interacción in vitro de células mononucleares con dos cepas de Staphylococcus aureus diferenciadas genotípica y fenotípicamente, aisladas de infecciones intramamarias (IIM) persistentes (P) y no persistentes (NP); y examinar su rol in vivo en un modelo de IIM experimental. Los estudios in vitro demostraron que los linfocitos Tγδ se activan tras el contacto con S. aureus, particularmente frente a la cepa 806 (NP), y que esta activación se potencia en presencia de otras poblaciones celulares. Asimismo, los linfocitos Tγδ contribuyen a la producción de IFN-γ e IL-17, mientras que TLR-2 no estaría involucrado en su activación. Además, los Tγδ modulan la activación de monocitos-macrófagos y la expresión de moléculas asociadas a la presentación antigénica de manera dependiente de la cepa. En el modelo in vivo, la cepa 806 (NP) indujo una respuesta inflamatoria temprana y eficaz, asociada a cambios en la frecuencia y el estado de activación de los linfocitos Tγδ, aumento de esta población en la leche y niveles elevados de IFN-γ e IL-17. En contraste, la cepa 5011 (P) mostró mayor capacidad para evadir estos mecanismos inmunes, favoreciendo el establecimiento de una infección persistente. Estos resultados demuestran que los linfocitos Tγδ participan en la respuesta inmune frente a S. aureus, y que las características genotípicas y fenotípicas de las cepas influyen en su comportamiento funcional, aportando información relevante para el diseño de estrategias de control de la mastitis bovina.
The aim of this thesis was to characterize the γδ T lymphocyte population following the in vitro interaction of mononuclear cells with two Staphylococcus aureus strains differing in their genotypic and phenotypic characteristics and isolated from persistent (P) and non-persistent (NP) intramammary infections (IMI), and to examine their role in vivo in an experimental IMI model. In vitro studies demonstrated that γδ T lymphocytes become activated upon contact with S. aureus, particularly in response to strain 806 (NP), and that this activation is enhanced in the presence of other immune cell populations. Moreover, γδ T lymphocytes contribute to IFN-γ and IL-17 production, whereas TLR-2 does not appear to be the main receptor involved in their activation. In addition, γδ T cells modulate monocyte–macrophage activation and the expression of antigen presentation–associated molecules in a strain-dependent manner. In the in vivo model, strain 806 (NP) induced an early and effective inflammatory response, which was associated with changes in the frequency and activation status of γδ T lymphocytes, an increase in this population in milk, and elevated local levels of IFN-γ and IL-17. In contrast, strain 5011 (P) exhibited a greater capacity to evade these immune mechanisms, thereby promoting the establishment of a persistent infection. Taken together, these results demonstrate that γδ T lymphocytes participate in the immune response to S. aureus, and that the genotypic and phenotypic characteristics of the strains influence their functional behavior, providing relevant information for the design of strategies to control bovine mastitis.
